I seem to be able to print shipping labels without having to "transmit" the manifest. How does that work? The shipping label I printed is not billed or valid until the manifest is sent?

Transmitting the manifest is only available to contract customers. Non-contract customers will use this simply as a group/categorizing feature for Canada Post shipments. More information can be found on the product page "Installation Instructions".
